In my microbiology lab I grew Staphylococcus epidermidis on my Mannitol Salt Agar (MSA) plate by inoculating the agar with a sample containing the bacteria and incubating it at an appropriate temperature for growth. MSA is both a selective and differential medium. To confirm the identification, I performed a Gram stain, which showed Gram-positive cocci, and a catalase test, which was positive. I identified the bacteria as S. epidermidis because the colonies grew but did not turn the medium yellow. This lack of yellow coloration indicates that the bacteria did not ferment mannitol, a key characteristic distinguishing S. epidermidis from other Staphylococcus species like S. aureus, which does ferment mannitol and turns the agar yellow.
